Specific allergen immunotherapy (SIT) is a unique therapy for allergic rhinitis because it provides symptomatic relief while modifying the allergic disease by targeting the underlying immunologic mechanisms. Sublingual (SLIT) is the most commonly prescribed routes for administering SIT. SLIT has been suggested to be a highly effective route of desensitization against a variety of allergens. In our prospective study we examined those people, who are allergic to ragweed. They keep a symptome diary, and we measurment for mechanism of immunological cells in their blood, before SLIT, and under and after the pollen season. The immunologycal cells visible, when we exemined a flow cytometry (FACS) by biomarkers. FACS is a powerful technique for analysis of multiple parameters of individual cells whithin heterogenous populations. Understanding mechanisms of inducible antigen-specific tolerance will improve immunotherapy and reveal the most relevant biomarkers for objectively measuring response to immunotherapy in clinical trials and routine practice. Various physiological and laboratory parameters are proposed as biomarkers of an immunological response to vaccines, although their surrogacy for clinical end points is unproven.

We look for a correlation between an objective and subjective outcome and a clinical endpoints, with statistical analysis and applied Bayesian-network. The first target is that, in the blood samples we found a biomarkers, which mark foreshadowe a clinical and immunological endpoints of SLIT, and an immunological modification of SLIT. These biomarkers base an decision supporting system the utility, efficiency interest for therapy.
